Dysfunction of autophagy and disturbed protein homeostasis are linked to the pathogenesis of human neurodegenerative diseases and the modulation of autophagy as the protein clearance process has become one key pharmacological target. Sigma 1 receptora ietekme uz peļu mezenhimālo stromas šūnu osteodiferenciāciju

Mezenhimālajām stromas šūnām (MSŠ) piemīt osteodiferenciācijas potenciāls. Sigma-1 receptors (SigmaR1) ir transmembrānu proteīns, tā ietekme uz osteodiferenciāciju nav vēl zināma. Bakalaura darba mērķis bija noskaidrot SigmaR1 ietekmi uz peļu taukaudu MSŠ (T-MSŠ) osteodiferenciāciju. Osteodiferenciācijas norise un SigmaR1 ietekme uz osteodiferenciāciju tika analizēta, pētot osteoģenēzē iesaistīto gēnu ekspresiju peļu T-MSŠ ar qPCR metodi. Salīdzinot izslēgta SigmaR1 (S1R-/-) T-MSŠ pret WT (wild type) T-MSŠ gēnu ekspresiju pēc osteodiferenciācijas, transkripcijas faktora osterix ekspresija tika nozīmīgi palielināta, bet nediferencētā stāvoklī būtiskas izmaiņas netika novērotas.

Ādas mezenhimālo cilmes šūnu fenotipa raksturojums pēc neirotrofo faktoru inducētas neirodiferenciācijas

Ādas mezenhimālās cilmes šūnas (Ā-MCš), pakļautas specifiskiem faktoriem, spēj diferencēties par neironiem un glijas šūnām. Maģistra darba mērķis bija analizēt Ā-MCš fenotipu pēc neirotrofo faktoru inducētas neirodiferenciācijas. Neiroektodermas gēnu ekspresiju Ā-MCš analizēja ar qPCR, cilmes šūnu pluripotences un mezenhimālo marķieru ekspresiju ar plūsmas citometriju, Sigma-1 receptora ekspresiju (SigmaR1) ar qPCR un IF, kā arī BDNF sekrēciju ar ELISA. Sigma-1 receptora gēna ietekme uz peļu taukaudu šūnu diferenciāciju par adipocītiem

No taukiem iegūtas mezenhimālās stromas šūnas (AdMSŠ) spēj diferencēties par adipocītiem. Pateicoties diferenciācijas potenciālam, tām ir plašs terapeitisko pielietojumu loks reģeneratīvajā medicīnā. Bakalaura darba mērķis bija izpētīt Sigma-1 (S1R) receptora gēna ietekmi uz peļu taukaudu šūnu diferenciāciju par adipocītiem in vitro. Lai analizētu adipodiferenciāciju AdMSŠ un S1R nozīmi, tika pārbaudīts četru ar taukaudiem saistītu gēnu CEBPA, PPARG, FABP4 un Adiponectin ekspresijas līmenis, kas tika analizēts ar qPCR. Neurodegenerative diseases are linked to dysfunctional proteostasis and disturbed autophagy. Here, we discuss how the sigma-1 receptor (Sig-1R) may act at the intersection of this interaction, as loss-of-function mutations of this unique chaperone are associated with defective autophagy and its pharmacological activation induces autophagic activity.

Background and Purpose Here, we describe the in vitro and in vivo effects of (4R,5S)-2-(5-methyl-2-oxo-4-phenyl-pyrrolidin-1-yl)-acetamide (E1R), a novel positive allosteric modulator of sigma-1 receptors. Taukaudu mezenhimālo stromālo šūnu iegūšana un raksturošana no SigmaR1 -/- pelēm

Maģistra darba mērķis bija veikt mezenhimālo cilmes (stromālo) šūnu (MCŠ) izdalīšanu no Sigma-1 receptora (SigmaR1) inaktivētā gēna peļu taukaudiem un noteikt to adipodiferenciācijas potenciālu. Šūnu fenotips tika raksturots, nosakot marķieru ekspresiju ar plūsmas citometriju, savukārt izdalīto šūnu adipodiferenciācija tika noteikta ar attēlu metodi un Oil-Red-O ekstrakciju. Fenotipēšana parādīja, ka lielākā daļa šūnu pieder pie MCŠ, ekspresējot CD29, CD44 un SCA-1 virsmas marķierus virs 90 % populācijas līmenī un daļēji ekspresējot CD105.
